PMS2 is a mismatch repair protein with a molecular weight of 96kDa. The PMS2 protein forms a heterodimer with the MLH1 protein and is then activated in the presence of ATP; this complex coordinates the binding of other proteins that repair DNA errors that occur during cell division. Loss of PMS2 expression in tumor cells helps identify Hmlh1 mutation vectors and determine whether they are suitable for mutation analysis. Defects in the PMS2 gene are responsible for a small but significant proportion of colorectal cancers and a majority of tumors with microsatellite instability. PMS2 has been linked to cases of the dominant genetic disorder hereditary nonpolyposis colon cancer (HNPCC) and, more clearly, to a variant hereditary nonpolyposis colon cancer called Turcot syndrome. PSM2 is often used in cancer research together with MLH1, MSH2 and MSH6.
