HRP/Alexa 488/Cy3 Tunel Cell Apoptosis Detection Kit
50T
The breakage of chromosomal DNA in cell apoptosis is a gradual and staged process. Chromosomal DNA is first degraded into large fragments of 50-300 kb under the action of endogenous nucleolytic enzymes, and then about 30% of the chromosomal DNA is degraded between nucleosome units under the action of Ca2+ and Mg2+-dependent endonucleases. are randomly cut off to form 180-200 bp nucleosomal DNA polymers. Therefore, in the late stage of apoptosis, DNA will be degraded into fragments of 180-200 bp, and a large number of 3′-OH ends are exposed on the broken genomic DNA. Terminal Deoxynucleotidyl Transferase (TdT) is a template-independent DNA polymerase that can catalyze the binding of deoxyribonucleotides to the 3′-OH end of broken DNA molecules. Therefore, the TUNEL (TdT mediated dUTP Nick End Labeling) apoptosis detection kit can be used to detect the fragmentation of nuclear DNA in tissue cells during the late process of apoptosis. The principle is that under the action of TdT enzyme, the 3´-OH end exposed when genomic DNA is broken is incorporated into HRP/Alexa 488/Cy3-dUTP, which can be detected by fluorescence microscopy or DAB color development method. This kit has a wide range of applications and is suitable for apoptosis detection in paraffin tissue sections, frozen tissue sections, cell slides, cell smears, etc.
