Nucleophosmin (NPM) is a protein encoded by the NPM1 gene in humans. The NPM1 gene is upregulated, mutated, and chromosomally translocated in a variety of tumor types. NPM1 chromosomal variation can be found in patients with non-Hodgkin lymphoma, acute promyelocytic leukemia, myelodysplastic syndrome, and acute myelogenous leukemia. NPM1 is usually overexpressed in solid tumors, and NPM1 is thought to promote tumor growth by inactivating the tumor suppressor p53/ARF pathway; in contrast, when NPM1 expression levels are low, NPM1 blocks tumor growth by inhibiting centrosome duplication.
