Uroplakin III is an asymmetric unit 47KDa glycoprotein. The protein can develop color on the surface of urothelial cells in formalin-fixed renal pelvis, ureter, bladder, and urethra tissues. This marker has only moderate sensitivity but high specificity for tumors of urothelial cell origin. Approximately 60% of human transitional epithelial cell-derived tumors (primary and metastatic) will express Uroplakin III protein focally (sometimes the expression may be minimal). In tumors derived from urothelial cells, loss of Uroplakin III protein is usually associated with high-grade tumors, invasion of myometrium, and lymphatic metastasis. Research data shows that Uroplakin III can be specifically expressed in p63-negative urothelial tumors, including undifferentiated carcinomas; it can also be expressed in vulvar Paget’s disease related to urothelial carcinoma.
