Xp11 translocation renal cell carcinoma is a newly discovered subtype of renal cell carcinoma that is more likely to occur in children and adolescents than in adults. Morphologically, these tumors show papillary architecture and clear cytoplasm, often with psammoma bodies. TFE3 is the most specific marker for Xp11.2 translocation renal cell carcinoma, and the mechanism may be that the formation of fusion genes promotes the overexpression of TFE3. Alveolar soft tissue sarcoma (ASPS) is a rare soft tissue sarcoma of uncertain differentiation that usually occurs in the extremities of young patients. It is characterized by chromosome rearrangements on 17q25 and Xp11.2, resulting in the ASPSCR1-TFE3 fusion gene and abnormal transcription. This unusual chimeric transcription factor retains the N-terminal and DNA-binding domains encoded by TFE3, while the ASPSCR1-encoding portion may provide a segment that regulates gene expression. This super-activated transcription factor may induce the expression of a large number of molecules and contribute to the diagnosis, evolution and metastasis of ASPS.
